Introduction
Mycoplasma contamination is a very difficult problem in the process of cell culture. Mycoplasma contamination can seriously affect eukaryotic cells, leading to changes in cell parameters such as cell proliferation and virus susceptibility, making the experimental results unreliable.
Mycoplasma is by far the smallest and simplest microorganism that can replicate itself. Mycoplasma lacks a cell wall and mainly relies on the growth of eukaryotic cell membranes. In most cases, mycoplasma contamination has no symptoms. Mycoplasma contamination cannot be observed through a microscope, and it will not cause rapid pH change and turbidity of the cell culture solution. Therefore, mycoplasma contamination in cell culture can exist for a long time without being noticed.

Mycoplasma contamination can be detected in many ways. Microbiology culture is considered to be the most sensitive method and serves as a reference for other detection techniques. However, the culture of Mycoplasma usually takes 2-4 weeks and requires specific experimental conditions, and it is impossible to detect Mycoplasma with harsh culture conditions. Fluorescence microscopy research is often used to screen mycoplasma, but fluorescence staining requires extremely strong results analysis experience. Enzyme-linked immunosorbent assay (ELISA) method can be used to detect surface antigens of mycoplasma, but the sensitivity is usually low. The polymerase chain reaction (PCR) method has high sensitivity and can determine the type of mycoplasma, but it is easy to get false positive or false negative results due to improper operation.

MycoProbe â„¢ detects 16S rRNA of Mycoplasma, its sensitivity is comparable to PCR method, but the common problems of PCR method will not appear.
