PNAS and other researchers published new findings on genetic traits of cancer cells

Researchers from the Beijing Academy of Sciences, Harvard University and other departments published an article titled "Reproducible copy number variation patterns among single circulating tumor cells of lung cancer patients" and found that although there are differences in genetic characteristics, tumor cells in the blood The genetic variation in cancer cells of the same patient is similar.

The research results were published in the journal Proceedings of the National Academy of Sciences (PNAS). Corresponding authors of the article were the famous Chinese scientist Professor Xie Xiaoliang, as well as associate researchers Bai Fan and Wang Jie of Peking University. Among them, Professor Xie Xiaoliang is a Chinese American academician. He is one of the pioneers of single-molecule biophysical chemistry and coherent Raman scattering microscopic imaging. His research group has conducted experiments on in vitro experiments and dynamics of single-molecule biological systems in living cells. A lot of important achievements have been made, especially the single-molecule fluorescence microscopy technology, such as coherent Raman micro-imaging technology (CARS, SRS) and so on.

At the end of last year, Science listed the first six major scientific fields in 2013 that are worthy of attention are single-cell, single-molecule technology. As a leading scientist in this field, Professor Xie Xiaoliang once published an article that developed the MALBAC amplification technology, which achieved the first high Whole genome sequencing of single sperm with coverage.

In this study, the researchers used this MALBAC technology to successfully achieve full-genomic amplification and deep sequencing of CTC from a single circulating tumor cell in the peripheral blood of cancer patients. MALBAC technology has many advantages such as uniform genome amplification and high coverage, which allows researchers to accurately detect genome-wide gene copy number variations (CNVs) and single nucleotide variations (SNVs) at the single cell level.

By analyzing circulating tumor cells in patients with lung adenocarcinoma and small cell lung cancer, the researchers detected important single nucleotide mutations and insertions / deletions in proto-oncogenes and tumor suppressor genes that are closely related to cancer development.

This shows that although some genetic characteristics of tumor cells flowing in the bloodstream are different in the cells, the difference in the number of gene copies of tumor cells in the blood of a patient is similar, even among several lung cancer patients of the same type Showed similarity.

This is the first time that the highly consistent copy number variation pattern observed in CTC will change the traditional understanding of tumor heterogeneity, revealing that specific copy number variation plays an important role in tumor formation and metastasis.

In addition, Professor Xie Xiaoliang also proposed a new solution to the problem of focusing fluorescence excitation on single mammalian fluorescent protein imaging in living mammalian cells: reflected light-sheet microscopy (RLSM) , Biological translation).

This is a new type of fluorescence microscopy technique that uses selective planar light to pass through the nucleus of living mammalian cells and uses a small mirror to reflect an elliptical laser beam from the top at a ninety degree angle, resulting in a thin light layer that is incident parallel Imaging plane, close to the sample surface. The signal-to-noise ratio produced by this optical layer is superior to the previous illumination light source and can achieve single fluorescent protein imaging with a time resolution of 100 Hz.

Most women won't leave home without putting on your makeup. Maybe you are one of them, and you find that it is not easy to fit the makeup process into your busy morning routine. Now a makeup set helps. A makeup set can make the process easily by grouping together coordinating colors, tools, and products in a single container.
