**Experimental Principle:**
This kit employs the double-antibody sandwich ELISA method to quantify mouse ovalbumin-specific IgE (OVA-sIgE) in biological samples. The process begins with the microplate being pre-coated with a specific monoclonal antibody that recognizes OVA-sIgE. Once the sample is added, any OVA-sIgE present will bind to the immobilized antibody on the plate. Subsequently, a horseradish peroxidase (HRP)-labeled secondary antibody, also specific for OVA-sIgE, is introduced to form a complex of antibody-antigen-enzyme-labeled antibody. After thorough washing to remove unbound components, the substrate TMB is added. Under the catalytic action of HRP, TMB turns blue, and when an acidic stop solution is applied, it changes to yellow. The intensity of the yellow color is directly proportional to the concentration of OVA-sIgE in the sample. A microplate reader is then used to measure the optical density (OD) at 450 nm. By comparing the OD values of the test samples with those of a standard curve, the exact concentration of mouse OVA-sIgE can be accurately determined. This method ensures high specificity and sensitivity, making it ideal for research applications involving allergic responses or immune system analysis.
