**Human Placental Protein 13 (PP13) Enzyme-Linked Immunosorbent Assay (ELISA) Kit – Instructions for Use**
This reagent is intended for research use only.
**Purpose:**
The Human Placental Protein 13 (PP13) ELISA Kit is designed to quantitatively measure the concentration of PP13 in human serum, plasma, urine, and other biological fluids. This assay is ideal for scientific studies related to placental function, pregnancy monitoring, and disease biomarker analysis.
**Principle of the Assay:**
This kit utilizes the double-antibody sandwich ELISA method. A microplate is pre-coated with a specific anti-PP13 monoclonal antibody. After adding the sample, PP13 binds to the immobilized antibody. A horseradish peroxidase (HRP)-labeled secondary antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following a washing step, the TMB substrate is introduced, leading to a color change that is proportional to the amount of PP13 present. The reaction is stopped with an acidic solution, turning the color from blue to yellow. The absorbance at 450 nm is measured using a microplate reader, and the concentration of PP13 is determined by comparing the sample’s OD value to a standard curve.
**Kit Components:**
- Microtiter Plate: 48 or 96 wells (coated with anti-PP13 antibody)
- Standard: 180 μg/L (0.5 mL × 1 bottle)
- Standard Diluent: 1.5 mL × 1 bottle
- Enzyme Conjugate: 3 mL × 1 bottle
- Sample Diluent: 3 mL × 1 bottle
- TMB Substrate (A & B): 3 mL × 1 bottle each
- Stop Solution: 3 mL × 1 bottle
- Wash Buffer (20×): 20 mL × 1 bottle (for 20–30 dilutions)
- Sealing Film: 2 pieces (for 48-well), 2 pieces (for 96-well)
- Storage: All components should be stored at 2–8°C.
**Sample Preparation Guidelines:**
- **Serum:** Collect blood, allow it to clot at room temperature for 10–20 minutes, then centrifuge at 2000–3000 rpm for 20 minutes. Store supernatant at 2–8°C.
- **Plasma:** Use EDTA or sodium citrate as anticoagulant. Mix well, centrifuge, and collect supernatant.
- **Urine:** Centrifuge at 2000–3000 rpm for 20 minutes.
- **Cell Culture Supernatant:** Centrifuge to remove debris before testing.
- **Tissue Samples:** Homogenize in PBS, centrifuge, and collect supernatant.
**Important Notes:**
- Avoid repeated freeze-thaw cycles.
- Do not use samples containing NaN3, as it may inhibit HRP activity.
- Always prepare a standard curve with duplicate wells for accurate results.
- Ensure all reagents are brought to room temperature before use.
- Discard any unused portions after opening.
**Procedure Summary:**
1. Prepare standard dilutions (120, 80, 40, 20, 10 μg/L).
2. Add sample and standards to designated wells.
3. Incubate at 37°C for 30 minutes.
4. Wash the plate 5 times with diluted wash buffer.
5. Add enzyme conjugate and incubate again.
6. Add TMB substrate and incubate for 15 minutes.
7. Stop the reaction with stop solution.
8. Measure OD at 450 nm and calculate concentrations using the standard curve.
