Professor Cheng Peng from Sun Yat-sen University published new results in stem cell research

In a groundbreaking study, researchers from Sun Yat-sen University have uncovered the potential of testicular Leydig stem cells (SLCs) as a promising cell source for treating testosterone deficiency in men. The findings were published in the November 21 issue of *Cell Research*, marking an important step forward in male hormonal health research. The lead author of the study is Professor Andy Peng Xiang from the Zhongshan School of Medicine at Sun Yat-sen University. A renowned expert in stem cell biology and developmental genetics, he has dedicated his career to understanding gene regulation, stem cell differentiation, and transgenic animal models. To date, he has contributed to over 30 scientific publications, showcasing his deep expertise in this field. Testosterone is a vital hormone for males, influencing numerous physiological processes throughout life—from fetal development to aging. It plays a key role in gender differentiation, physical growth, and overall body function. Beyond the biological, it also affects mood, behavior, and social interactions. Testosterone deficiency is a common condition that can occur at any age, leading to issues such as reduced muscle mass, fatigue, sexual dysfunction, and cognitive decline. Leydig cells are responsible for testosterone production in the testes, playing a critical role in male fertility and secondary sexual characteristics. However, current treatments like Leydig cell transplantation face limitations—mature Leydig cells cannot divide, have short survival times, and often fail to maintain long-term function, reducing their therapeutic effectiveness. Like other specialized cells, Leydig cells originate from undifferentiated stem cells known as SLCs. These stem cells continuously replenish the Leydig cell population through self-renewal and differentiation. Due to their ability to produce testosterone and their compatibility with the body’s natural regulatory systems, SLCs are considered a highly promising candidate for cell-based therapies. However, isolating and purifying them has been challenging due to the lack of specific markers. In this latest study, scientists used flow cytometry combined with functional assays to identify and isolate SLCs based on Nestin (Nes) expression. Using a mouse model where Nes drives GFP expression, they found that Nes-GFP+ cells localized in the interstitial compartment, matching the location of SLCs. These cells expressed LIFR and PDGFR-α but not typical Leydig cell markers. Further experiments showed that these cells could form clones, self-renew, and differentiate into multiple lineages, including Leydig cells, while producing testosterone. When transplanted into mice with damaged or aged Leydig cells, the Nes-GFP+ cells successfully integrated into the testicular stroma, boosting testosterone levels and promoting germ cell recovery. The study also suggests that CD51 may serve as another surface marker for SLCs, offering new tools for future research and clinical applications. This discovery opens up exciting possibilities for developing more effective and sustainable treatments for testosterone deficiency, potentially improving the quality of life for millions of men worldwide.
